Prenatal methylmercury exposure and DNA methylation in seven-year-old children in the Seychelles Child Development Study

چکیده
Methylmercury (MeHg) is present in fish and a neurotoxicant at sufficiently high levels. One potential mechanism of MeHg toxicity early life epigenetic dysregulation that may affect long-term neurodevelopment. Altered DNA methylation nervous system-related genes has been associated with adult mental health outcomes. To assess associations between prenatal exposure (at the cytosine CG dinucleotides, CpGs) three genes, encoding brain-derived neurotropic factor (BDNF), glutamate receptor subunit NR2B (GRIN2B), glucocorticoid (NR3C1), children who were exposed to utero. We tested 406 seven-year-old Seychellois participating Seychelles Child Development Study (Nutrition Cohort 2), prenatally from maternal consumption. Total mercury hair (prenatal measure) collected during pregnancy was measured using atomic absorption spectroscopy. Methylation children’s saliva by pyrosequencing. CpG seven years age, we used multivariable linear regression models adjusted for covariates. identified one GRIN2B two NR3C1 CpGs out 12 total sites. Higher higher each site. For example, CpG3 had an expected increase 0.03-fold additional 1 ppm (B = 0.030, 95% CI 0.001, 0.059; p 0.047). Several sites are located transcription binding observed changes predicted lead lower gene expression. In population people consume large amounts fish, showed differential age specific influence neurodevelopment health.

Prenatal methylmercury exposure from ocean fish consumption in the Seychelles child development study.
INTRODUCTION Exposure to methylmercury (MeHg) before birth can adversely affect children's neurodevelopment. The most common form of prenatal exposure is maternal fish consumption, but whether such exposure harms the fetus is unknown. We aimed to identify adverse neurodevelopmental effects in a fish-consuming population. METHODS We investigated 779 mother-infant pairs residing in the Republic...
متن کاملSemiparametric modeling of age at achieving developmental milestones after prenatal exposure to methylmercury in the Seychelles child development study.
Controversy exists concerning the fetal risk associated with exposure to low-dose methylmercury from maternal fish consumption. Previous studies of the effects of acute prenatal mercury exposure identified delays in achieving developmental milestones among exposed children. This led to public health concern that prenatal low-dose exposure from fish consumption could adversely affect the fetus. ...

INTRODUCTION People worldwide depend upon daily fish consumption as a major source of protein and other nutrients. Fish are high in nutrients essential for normal brain development, but they also contain methylmercury (MeHg), a neurotoxicant. Our studies in a population consuming fish daily have indicated no consistent pattern of adverse associations between prenatal MeHg and children's develop...
متن کاملAssociations of Baroreflex Sensitivity, Heart Rate Variability, and Initial Orthostatic Hypotension with Prenatal and Recent Postnatal Methylmercury Exposure in the Seychelles Child Development Study at Age 19 Years
BACKGROUND A few studies have suggested an association between prenatal exposure to methylmercury and decreased heart rate variability (HRV) related to autonomic heart function, but no study has examined this association using baroreflex sensitivity (BRS). In this study we assessed the distribution of BRS and immediate orthostatic hypotension (IOH) in young Seychellois adults and their associat...
